Identification of Cells Induced to Synthesize Hemoglobin
نویسنده
چکیده
In order to facilitate the identification of bone marrow cells in which hemoglobin synthesis is initiated, erythropoiesis was first suppressed in guinea pigs through the induction of posthypoxic polycythemia, and then it was restimulated by bleeding and reexposure to hypoxia. Hemoglobin synthesis was detected with “Fe incorporation on radioautographs, and its presence was demonstrated in the light microscope with the benzidine reaction and absorption of monochromatic light at A 4046 A. In the electron microscope, hemoglobin was detected in the cytoplasm by a general increase in electron density after treating the tissue with diaminobenzidine (DAB) and 0s04. Densitometric measurements were carried out on electronmicroscopic negatives, using reticular cell cytoplasm as a base line. In normal marrow, proerythroblasts were the earliest cells in which hemoglobin could be detected, but during the early phase of erythropoietic stimulation, hemoglobin was demonstrated in transitional cells with all the methods employed. Without the specific demonstration of hemoglobin, these cells could not be recognized morphologically as erythroblasts nor could they be distinguished from the precursors of bone marrow small lymphocytes. Transitional cells were numerous in the marrow at the time of stimulation, and 40 hr later a small number of them were labeled with “Fe and synthesized hemoglobin in detectable amounts. Proerythroblasts were absent at the time of the stimulus, and when they reappeared the majority were benzidine or DAB positive and had incorporated “Fe. The findings suggest that progenitor cells of erythroblasts are among the basophilic members of the transitional cell population, and erythropoietic stimulation induces hemoglobin synthesis in them. The relationship of these cells to the progenitors of other hemopoietic cells, as well as to the pluripotent stem cell, is discussed.

متن کاملOn the mechanism of erythropoietin-induced differentiation. IX. Induced synthesis of 9 S ribonucleic acid and of hemoglobin.
Erythropoietin induces the synthesis of 9 S RNA and hemoglobin by cultured bone marrow cells derived from polycythemic rats and mice in which hemoglobin synthesis has been suppressed. Synthesis of 9 S RNA starts within 1 hour (rat cells) and 4 hours (mouse cells) of addition of erythropoietin to the medium, while hemoglobin synthesis is not initiated until 8 to 10 hours later. These findings in...

Age related cytochemical changes of thymic endocrine cells were studied in 78 day old chicks at five day interval to age of day 60 employing a panel of cytochemical stains. Methenamine silver revealed cell morphology including cell processes distinctly while diamine silver revealed a stronger argentaffinity in these cells. The cells had greater affinity for diamine silver compared to methenamin...

Coenzyme Q 10 is a natural antioxidant and free radicals scavenger. In the present study, we examined effect of coenzyme Q 10 on hemoglobin A 1C , serum urea and creatinine in alloxan-induced Type 1 diabetic rats. Thirty Sprage-Dawley male rats were divided into three groups randomly; group one as control, group two diabetic untreatment, and group three treatments with coenzyme Q ...
متن کاملEffect of Oxidized Low Density Lipoprotein on the Expression of Runx2 and SPARC Genes in Vascular Smooth Muscle Cells
Background: Vascular calcification is an important stage in atherosclerosis. During this stage, vascular smooth muscle cells (VSMC) synthesize many osteogenic factors such as osteonectin (encoded by SPARC). Oxidative stress plays a critical role in atherosclerosis progression, and its accumulation in the vascular wall stimulates the development of atherosclerosis and vascular calcification. The...
